Isolation of RNA; A new guanidine thiocyanate procedure for the isolation of full-length RNA templates for reverse transcription from tissues rich in RNase and proteoglycans.
用途：离液剂和强效变性剂;溶解细胞。用于变性裂解细胞；提取RNA和DNA;无RNA酶和DNA酶活性。TRIzol是一种新型总RNA抽提试剂，可以直接从细胞或组织中提取总RNA。其含有苯酚、异硫氰酸胍等物质，能迅速破碎细胞并抑制细胞释放出的核酸酶。
异硫氰酸胍法提取制备模板核酸此法主要用于RNA的提取.标本为组织细胞及血清等.
1.异硫氰酸胍消化液
异硫氰酸胍4mol/L
柠檬酸钠(PH7.0)25mmol/L
十二烷基肌酸钠0.5%
β-巯基乙醇0.1mol/L
2.消化方法：用50～100ul细胞悬液及血清，加等体积的异硫氰酸胍消化液振混匀后，或65℃1小时，或室温放置数分钟，然后加1/10体积的3mmol/LpH5.2的醋酸钠缓冲液，再加等体积的酚：氯仿，用力振摇约10s，10000r/min，离心5min，取上清加等体积异丙醇-20℃放置3h后，14000r/min离心15min，沉定用75%冰冷乙醇离心洗涤1～2次，真空干燥，沉淀用TE缓冲液溶解即可用于逆转录PCR扩增，或放-20℃保存。
