Biochem/physiol Actions
Tobramycin is an aminoglycoside.
Mode of Action: Binds to 70S ribosomal subunit; inhibits translocation; elicits miscoding.
Spectrum of Activity: Gram negative bacteria. Not effective against Enterococci.
Tobramycin binds irreversibly to one of two aminoglycoside binding sites on the 30 S ribosomal subunit, inhibiting bacterial protein synthesis. Tobramycin may also destabilize bacterial membranes by binding to 16 S r-RNA. It inhibits translocation and elicits miscoding.
Application
Tobramycin is an aminoglycoside, broad-spectrum antibiotic produced by Streptomyces tenebrarius. It is effective against gram-negative bacteria, especially the pseudomonas species. It is not effective against Enterococci. It is used to inhibit bacterial protein synthesis. It is used to treat pseudomonas aeruginosa lung infections and is used in combination with other antibiotics to treat urinary tract infections, gynecologic infections, peritonitis, endocarditis, pneumonia, sepsis, respiratory infections, osteomyelitis, and other soft-tissue infections. It is a potential therapy for sinus infections.
妥布霉素能够联结在30S和50S的联结位置,阻碍70S复合物的形成,使mRNA不能翻译成蛋白质,导致细胞死亡。主要对革兰阴性菌,如绿脓杆菌、大肠杆菌、克雷白杆菌、肠杆菌属、变形杆菌、枸橼酸杆菌有效。临床主要用于敏感细菌引起的严重感染,如革兰阴性菌特别是绿脓杆菌、大肠杆菌及肺炎杆菌等引起的烧伤感染、败血症、呼吸系统感染、泌尿系统感染、胆囊胆道感染及软组织严重感染等。
