Polymyxin B destabilizes and disrupts cell membrane integrity which allows the leakage of cellular molecules and inhibits cellular respiration. In human dendritic cells, DCs incubated with polymyxin B up-regulate the expression of HLA class I and II, the co-stimulatory CD86 molecule, and show an increase in the fraction of adherent cells.In the rat model, Polymyxin B caused a significant reduction in plasma endotoxin and tumour necrosis factor α (TNF-α) concentration.

Application
Polymyxin B is a lipopolysaccharide binding antibiotic derived from Bacillus polymyxa. It is primarily used against Gram-negative bacteria. It is used to mature dendritic cells in culture and inactivate potential lipopolysaccharide contaminations . Polymyxin B is used to study potential treatments for septic shock and for antimicrobial susceptibility testing of organisms such as Pseudomonas aeruginosa. 硫酸多粘菌素B (多粘菌素B) 英文名：Polymyxin B Sulfate (Aerosporin)多粘菌素系由多粘芽胞杆菌（bacilluspolymyxa）产生的一组多肽类抗生素。多粘菌素b和e供药用。常用其硫酸盐，为白色结晶性粉末，易溶于水，有引湿性。在酸性溶液中稳定，其中性溶液在室温放置一周不影响效价，碱性溶液不稳定。
其抗菌谱及临床应用与多粘菌素E相似，对革兰阴性杆菌，如大肠杆菌、绿脓杆菌、副大肠杆菌、肺炎克雷白杆菌、嗜酸杆菌、百日咳杆菌及痢疾杆菌等有抑制或杀菌作用。
